Dupilumab, which is currently in Phase III of development for eosinophilic asthma by Regeneron and Sanofi, is a fully-humanized mAb against the interleukin-4 receptor a subunit (IL-4-Ra) that inhibits both the IL-4 and IL-13 signaling pathways. The similarities in activity between IL-13 and IL-4 in asthma pathogenesis have been established (Steinke et al., 2001; Wong et al., 2001). IL-4 is functionally similar to IL-13, and is present in increased levels in the sputum and bronchial tissue of asthmatic patients. It seems plausible that an agent that inhibits the activity of both IL-4 and IL-13 would be more effective (Wong et al., 2001). Therefore, dupilumab could have a ripple effect on the sales of IL-13 inhibitors, lebrikizumab, and tralokinumab.
